Asensio sealed victory for Madrid in the 70th minute with a curling shot from the edge of the penalty area which deflected off a defender and wrong-footed the goalkeeper before going into the net.
Madrid manager Carlo Ancelotti handed a first LaLiga start since September to Eden Hazard as he made a number of changes to his lineup ahead of their trip to Manchester.
Madrid and Manchester City drew 1-1 in the first leg of the semifinal in Madrid.
